In vitro Propagation of Mabolo (Diospyros discolor Willd.) Using Varying Concentrations of Naphthalene Acetic acid NAA Benzyladenine BA

Abstract

In vitro propagation of Mabolo (Diospyros discolor Willd.) was conducted to establish sterilization procedure using Knudson C medium with varying concentrations of Naphthalene acetic acid (NAA) and Benzyladenine (BA). The following treatment concentrations were used: TO (control, Knudson C only), T1 (Knudson C + 0.5mg / L NAA + 0.5mg / L BA), T2 ( Ison C + 1mg / L * NAA + 1mg / L * BA ), T3 (Knudson C + 2 mg/ LNAA + 2mg / L BA). One hundred percent (100%) in vitro germination was obtained in all treatments 3 weeks from inoculation. Sterilization procedure for this plant species was established using dehusked seeds with tegmen removed. Based on the developmental stages of in vitro germinated seeds, T2 was considered as the best treatment since two primary roots were developed after the elongation of hypocotyl.
